Divalent ions do inhibit trypsin activity. edta is used to chelate free magnesium and calcium ions in order to maintain the inhibition of trypsin activity. It is recommended that cells be washed with EDTA prior to trypsin treatment to eliminate all divalent ions from the culture medium.
