Carrageenan-Induced Air Pouch Synovitis Model

The subcutaneous air pouch is a model used to study acute inflammation in vivo. It was first described by Selye in 1953. The pouch is created by injecting sterile air into the intra-scapular area of the rat or mouse's back. Later, various irritants can be injected into the pouch to induce inflammation. The lining of the pouch is composed primarily of macrophage-like (Type A) and fibroblast-like (Type B) cells, similar to the synovial cavity. Injection of a carrageenan solution into the pouch triggers an inflammatory reaction characterized by cell infiltration, increased exudate, and the production of pro-inflammatory mediators, including prostaglandins, leukotrienes, cytokines, and oxidative stress response components.

After acclimation, sterile air is injected into the subcutaneous tissue at the midline of their backs. Following this, a solution of carrageenan in sterile saline is injected into the animals' air pouch.
