Droplet Digital PCR (ddPCR) Service
Droplet Digital PCR (ddPCR) is a great technology that provides ultrasensitive nucleic acid detection and absolute quantification.
The ddPCR method involves dividing nucleic acid samples into numerous microdroplets of known volume within a water-oil emulsion. Amplification of specific nucleic acid sequences occurs individually within each microdroplet using the same process and materials typically employed in a standard TaqMan® probe-based assay. Following amplification, fluorescence detection is used to categorize each droplet as either PCR-positive or PCR-negative. The concentration of target DNA/RNA templates (copies/µl) in the initial sample is then determined using a Poisson distribution based on the counted droplets.

ddPCR can be performed as a standalone analysis or combined with our Transgene Mapping (Locus Amplification & Sequencing) assays, which provide comprehensive information about the transgene vector, including the vector copy numbers, vector integration sites, and vector integrity.
ddPCR can also be used to do Pre-MCB Clone Check Clone Characterization. When scaling up production, cells sometimes show a diminished growth rate. This can be the result of a high vector copy number. Excessive copies can create a metabolic burden on the cells resulting in stress, imbalanced gene expression, and viability issues.
