Partial Sciatic Nerve Ligation (PSL/PSNL) Model

The management of chronic pain continues to be a significant challenge, with existing treatments often accompanied by undesirable side effects such as tolerance and addiction. Chronic neuropathic pain arises from damage or disease within the somatosensory system. In the quest for therapies with fewer adverse effects, animal models of pain are indispensable in preclinical research. The PSL/PSNL model is a method that generates chronic neuropathic pain in animals, manifesting as mechanical and thermal hypersensitivity, persistent pain, and alterations in limb temperature. This model is particularly well-suited for the preclinical study of neuropathic pain due to its ability to replicate many of the symptoms experienced by humans with this condition. Additionally, the surgical technique is relatively quick and simple to execute, making it a valuable tool in the investigation of neuropathic pain.

- Modeling Method
After anesthetization, an incision will be made in the skin and the muscle will be blunt dissected to expose the sciatic nerve. In the model group, the nerve is carefully freed and one-third to one-half of the nerve is tightly ligated with silk suture. In the sham group, the sciatic nerve is left intact and the muscle is sutured, and the skin closed with staples.
