Never can cryopreserved cells be kept at -20 °C.
Theoretically, there is no limit to the number of passages, but in practice, due to changes in the culture environment of the isolated culture, serum quality, medium quality, passaging damage and other external culture environment, the cells will have problems of differentiation, non-proliferation, aging and apoptosis if they are more than 10 generations old, so it is recommended to use them in less than 10 generations in regular practice.
