Never can cryopreserved cells be kept at -20 °C.
It is likely that the culture system is not suitable for the cells (the recommended culture system is not used); or the digestion is excessive, which has a serious effect on the cells, or the passaging ratio is inappropriate (cells with density-dependent properties are passaged too sparsely, or cells with faster growth are passaged more densely, and the cells grow in severe stacks).
