Never can cryopreserved cells be kept at -20 °C.
Firstly, we should check whether the isolation method is correct, and it is better to perform microscopic examination for each step in the middle, so that we can find out which step loses cells, and secondly, we should consider whether the later centrifugation step loses cells.
