When cells are frozen directly without any protective agent, water inside and outside the cells can quickly form ice crystals, which can cause a series of adverse reactions. If more ice crystals are formed inside the cell, with the decrease of freezing temperature, the volume of ice crystals expands causing irreversible damage to the DNA spatial conformation of the cell nucleus, which leads to cell death.
