Description
NAP-2 (CXCL7) is a CXC chemokine that can signal through the CXCR1 and CXCR2 receptors. It is produced in leukocytes by enzymatic processing of a precursor called platelet basic protein (PBP). NAP-2 chemoattracts and activates neutrophils. It is a protein that is released in large amounts from platelets following their activation. It stimulates various processes including mitogenesis, synthesis of extracellular matrix, glucose metabolism and synthesis of plasminogen activator. Recombinant rat NAP-2 produced in E. coli is a single, non-glycosylated polypeptide chain of 62 amino acids and a molecular mass of 6.8 kDa. It has been purified by proprietary chromatographic techniques.

Activity
NAP-2 is fully biologically active when compared to a standard. Biological activity was determined by its ability to chemoattract BaF3 mouse pro-B cells transfected with human CXCR2. The ED50 for this effect is less than 10 ng/ml, corresponding to a specific activity of Greater than1 x 10^5 units/mg.

Formulation
White (freeze-dried) powder. Lyophilized from a sterile-filtered (0.2 μm) concentrated protein solution in 1×PBS (pH 7.4).
Reconstitution
Please centrifuge product before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rRa NAP-2 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage, it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
