Description
Leukemia Inhibitory Factor also called LIF is a lymphoid factor that promotes long-term maintenance of embryonic stem cells by suppressing spontaneous differentiation. Leukemia Inhibitory Factor has several functions such as cholinergic neuron differentiation, control of stem cell pluripotency, bone & fat metabolism, mitogenesis of factor dependent cell lines & promotion of megakaryocyte production in vivo. Human and mouse LIF exhibit a 78% identity in its amino acid sequence. Human LIF is as active on human cells as is it is on mouse cells, though mouse LIF is about 1000 fold less active on human cells, than human LIF. Mouse recombinant Leukemia Inhibitory Factor (LIF) produced in E. coli is a single, non- glycosylated, polypeptide chain containing 181 amino acids and having a molecular mass of 20 kDa.

Activity
Activity of mouse LIF was determined by its ability to induce differentiation of murine M1 myeloid leukemic cells. The specific activity is Greater than 1 x 10^8 IU/mg, where 50 U is defined as the amount of murine LIF required to induce differentiation in 50% of the M1 colonies per ml of agar culture.

Formulation
Sterile-filtered, white, lyophilized (freeze-dried) powder. The protein was lyophilized from a concentrated (1 mg/ml) solution in 20 mM Phosphate buffer (pH 7.4) and 0.02% Tween-20.
Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultrapure water or aqueous buffers to a concentration of 0.1- 1.0 mg mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted protein should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
