Description
Interleukin 12 (IL-12) is a regulatory protein produced by activated B lymphocytes and macrophages. The biological activities of IL-12 include the stimulation of growth of activated CD4+ and CD8+T cells and NK cells. Human IL-12 also promotes the development of proinflammatory/TH1-like CD4+ cells and cytotoxic CD8+T cells. Murine IL-12 is a disulfide linked heterodimeric protein with a molecular weight of 75 kDa comprised of disulfide-bonded 35 kDA (p35) and 40 kDa (p40) subunits.

Activity
Determined by ist ability to stimulate the proliferation of 2D6 cells. The expected ED50 is ≤ 0.1 ng / ml, corresponding to a specific activity of Greater than 1 ×10^7 units/mg. The ED50 has also been determined by the dose-dependent stimulation of INF-gamma production by NK cells co-stimulated with IL-18 is ≤ 0.1 ng / ml, corresponding to a specific activity of Greater than 1 ×10^7 units/mg.

Less than 1 EU/μg.
Formulation
Lyophilized from a sterile-filtered protein solution in 1 x PBS (pH 7.2).
Reconstitution
Please centrifuge product before opening vial. Reconstitute in sterile PBS (pH 7.2) to a concentration of 0.1 - 1.0 mg/ml. Do not vortex. This solution can than be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted murine IL-12 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
