Description
Interleukin-1 receptor antagonist (IL-1 RA) is a member of the IL-1 family. Endogenous IL-1 RA is produced in numerous animal disease models as well as in human autoimmune and chronic inflammatory diseases. IL-1 RA is a naturally occurring protein which inhibits the activity of IL-1- alpha and IL-1-beta by competitively blocking binding to their type I and type II receptors. It binds to IL-1 receptors in competition with IL-1, but does not elicit intracellular response from this binding. Its role in counteracting the proinflammatory effects of IL-1 is being studied by numerous research groups. IL-4 and IL-13 have been shown to amplify the stimulatory effect of IL-1-beta on the production of soluble and intracellular forms of IL-1 RA. The regulated expression of IL-1 RA in various cell types has been shown to be influenced by cytokines. In synovial fibroblasts the synthesis of IL-1 RA is markedly enhanced by IL-1, TNF-alpha or PDGF. Recombinant mouse IL-1 RA produced in E. coli is a single, non-glycosylated polypeptide chain of 153 amino acids and a molecular mass of 17.4kDa. It has been purified by proprietary chromatographic techniques.

Activity
Recombinant Interleukin-1 RA is fully biologically active when compared to a standard. The ED50, as determined by the dose-dependant inhibition of IL-1 stimulation of D10S cells, was found to be 0.5 ng/ml corresponding to a specific activity of 2 x 10^6 units/mg.

Formulation
Lyophilized from a sterile-filtered protein solution with no additives.
Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted ru IL-1 RA should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
