Description
VEGF (Vascular endothelial growth factor) is the only growth factor that stimulates vascular permeability. It promotes endothelial proliferation and survival, angiogenesis, vasculognesis and inhibits apoptosis. VEGF mitogenic activity is specific for endothelial cell and that makes it distinct among other growth factors. VEGF is thought to be important in the pathophysiology of neuronal and other tumors, by functioning as a promter of angiogenesis for human gliomas. VEGF-121 has increased acidity which allowes it to circulate more freely than other VEGF forms. Biological activities of VEGF are not species-specific and glycosylation is not required for biological activity. Recombinant human VEGF-121 produced in plant contains 110 amino acids and a 16 a.a. histidine-based tag for a total length of 126 a.a., and has a predicted molecular mass of 14.4 kDa. As a result of glycosylation, the recombinant protein migrates with an apparent molecular mass of 19 kDa in SDS-PAGE.

Activity
The ED50 is determined by a cell proliferation assay measuring the dose-dependent effects of serial dilutions of human VEGF-121 on HUVEC cells. The ED50 value for VEGF121 lies typically in the range of 2.8-4.3 ng/ml. The optimal concentration should be determined for specific applications and cell lines.

Formulation
Sterile filtered through a 0.2 μm filter. Lyophilized from 10 mM acetic acid (pH 5.0).
Reconstitution
Please centrifuge the vial before opening. It is recommended to reconstitute the lyophilized protein in sterile water to a concentration of no less than 100 μg/ml. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted protein should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
