Description
BAFF (B lymphocyte activating factor) is a member of the tumor necrosis factor (TNF) ligand family which is expressed in T Cells, macrophages, monocytes and dendritic cells. It is also known as BLyS, THANK, TALL, zTNF4 and TNFS20. BAFF enhances B cell survival in vitro and has emerged as a key regulator of peripheric B cell and it is vital homeostatic cytokine for B cells that helps regulate both innate and adaptive immune responses. BAFF binds to three TNF receptors: B-cell maturation antigen (BCMA/TNFRSF17), transmembrane activator and calcium modulator and cyclophilin ligand interactor(TACI/TNFRSF13B) and BAFF receptor (BAFF R/BR3/TNFRSF 13C).The human BAFF gene code for a 285 amino acids type II transmembrane protein. Recombinant human soluble BAFF is a 151 amino acids containing the TNF-like portion of the extracellular domain of BAFF. Recombinant BAFF has been produced by transient expression of BAFF in non-transgenic plant. It consists of a glycosilated polypeptide chain with 151 amino acids (134-285 aa; Q9Y275 [TN13B_HUMAN]) fused to a 10XHis tag at the N-terminus and has a molecular mass betwen 18 and 20kDa. Recombinant BAFF has been purified by sequential chromatography (affinity and anionic FPLC) and does not contain animal–derived components or impurities.

Activity
The biological activity has been determined by the dose-dependent stimulation of the proliferation of B cells from human PBMC. Cell proliferation was measured using the MTT method. The ED50 was found to be ≤50 ng/ml*. *Activity results may vary with PBMC donors.

Formulation
Recombinant human BAFF is lyophilized from 20 mM PBS buffer (pH 7.0) and 0.2 M NaCl.
Reconstitution
Please centrifuge product before opening the vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 25-50 μg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted protein should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage, it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
