ActoFactor™ Recombinant Human Phospholipase A2, group VII (platelet-activating factor acetylhydrolase, plasma)

  • Specification
Cat.No.
CSC-CTK0471
Description
Platelet Activating Factor (PAF) is a biologically active phospholipid, which exerts primarily proinflammatory activities by specifically signaling through G-protein-coupled receptors on platelets, neutrophils, and monocytes. Platelet Activating Factor Acetylhydrolase (PAF-AH) is a secreted protein that mediates PAF activity by specifically catalyzing hydrolysis of the "sn2" ester bond, resulting in the conversion of PAF to the biologically inactive lyso-PAF. PAF-AH can also interact with LDL particles to induce the hydrolysis of LDL associated, oxidized phospholipids, generating lysophosphatidylcholine (lyso-PC) and other lysophospholipids. Recombinant PAF-AH is a 420 amino acid glycoprotein which migrates with an apparent molecular mass of 47-55 kDa by SDS-PAGE analysis.
Species
Human
Product Overview
Human PLA2G7 expressed in 293 cells
Molecular Mass
47 - 55 kDa
Size
CAT# CSC-CTK0471-20 (20 μg); CAT# CSC-CTK0471-100 (100 μg)
Expression System
293 cells
Purity
Greater than 95% as determined by SDS-PAGE and HPLC analysis.
Activity
Measured by its ability to cleave a PAF analog in a chromogenic substrate linked assay. At a PAF- AH concentration of 10.0 μg/ml, 50% cleavage was achieved at an incubation time of approximately 2 minutes.
Endotoxin Level
Less than 1 EU/μg.
Formulation
Lyophilized from a sterile-filtered solution with no additives.
Reconstitution
Please centrifuge product before opening vial. The lyophilized protein should be reconstituted with distilled, sterile water to a concentration of 0.1-1.0 mg/ml. For further dilution, carrier protein (0.1% BSA or HSA) should be added to avoid loss of bioactivity.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted protein should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
