ActoFactor™ Recombinant Human Fas ligand (TNF superfamily, member 6)
- Specification
Cat.No.
CSC-CTK0496
Description
Fas Ligand (FasL) is a member of the TNF superfamily that is expressed on the cell surface of activated T cells. Binding of FasL to Fas Receptor triggers apoptosis in Fas-bearing cells. FasL has the ability to kill T cells and activated B cells which leads to down-regulation of the immune response. The mechanism of Fas induced apoptosis involves recruitment of pro-caspase 8 through an adaptor molecule called FADD followed by processing of the pro-enzyme to active forms. These active caspases then cleave various cellular substrates leading to the eventual cell death. Both human and murine sFasL are fully active on human and murine cells. Recombinant human soluble Fas Ligand is a 17.9 kDa protein (156 amino acid residues) comprising the TNF homologous region of FasL and contains an 8 residue N-teminal His-Tag.

CAT# CSC-CTK0496-10 (10 μg); CAT# CSC-CTK0496-50 (50 μg)
Expression System
CHO cells
Purity
Greater than 95% as determined by SDS-PAGE and HPLC analysis.
Activity
Determined by its ability to induce cytotoxicity in Jurkat cells in the absence of any cross-linking. The ED50 for this effect is 1 x 10^5 units/mg.
Endotoxin Level
Less than 1 EU/μg.
Formulation
Lyophilized from a sterile-filtered solution in 10 mM sodium phosphate (pH 7.5) and 150 mM NaCl.
Reconstitution
Please centrifuge product briefly before opening the vial. Reconstitute in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized powder, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted sFasL should be used immediately or stored in undiluted working aliquots at -20°C. Avoid repeated freeze / thaw cycles.
