ActoFactor™ Recombinant Human Chemokine (C-X-C motif) ligand 9
- Specification
Cat.No.
CSC-CTK0066
Description
Monokine induced by Interferon-g (MIG) is a member of the a (CXC) family of chemokines. It is produced by lymphokine activated macrophages, IFN-g-treated peripheral blood monocytes, IFN-g-treated primary glial cells of the central nervous system. MIG is a chemoattractant for activated T lymphocytes and tumor-infiltrating lymphocytes but not for neutrophils or monocytes. The mature protein contains 103 amino acid residue and migrates with an apparent Mr of 11.7 kDa. MIG has an extended C-terminus containing greater than 50% basic amino acid residues. The C-terminus is prone to proteolytic cleavage resulting in size heterogeneity. MIG binds to CCR3.
Species
Human
Product Overview
Human CXCL9 expressed in E.coli
Sequence
A DNA sequence encoding the mature human MIG protein sequence (TPVVRKGRCSCISTNQGTIHLQSLKDLKQFAPSPSCEKIEIIIATLKNGVQTCLNPDSADVKELIKKEKQVSQKKKQKNGKKHQKKKVLK VRKSQRSRQKKTT) was expressed in E. coli.
Size
CAT# CSC-CTK0066-10 (10 μg); CAT# CSC-CTK0066-50 (50 μg)
Expression System
E.coli
Purity
Greater than 97% as determined by SDS-PAGE and visualized by silver stain
Activity
Activity was determined by ability to chemoattract human lymphocytes cultured in the presence of IL-2 for 14 - 21 days, or chemotaxis of CXCR3 transfected mouse BAF/3 cells. The ED50 for these effects were typically 0.2 - 0.6 μg/mL, or 1.5 - 8 μg/mL, respectively.
Endotoxin Level
Endotoxin level less than 0.1 ng per 1 μg of the cytokine as determined by the LAL method.
Formulation
Lyophilized from a 0.2 μm filtered solution in 30% CH3CN and 0.1% TFA containing 50 μg of bovine serum albumin per 1 μg of cytokine.
Reconstitution
It is recommended that sterile PBS containing at least 0.1% human serum albumin or bovine serum albumin be added to the vial to prepare a stock solution of no less than 20 μg/mL.
Storage & Stability
Lyophilized samples are stable for greater than six months at -20°C to -70°C. Reconstituted MIG/CXCL9 can be stored under sterile conditions at 2°C to 4°C for one month or at -20°C to -70°C for three months without detectable loss of activity. Avoid repeated freeze-thaw cycles.
