Description
BMP-2 belongs to the transforming growth factor-beta (TGF-b) superfamily. Bone morphogenic protein induces bone formation. BMP-2 is a candidate gene for the autosomal dominant disease of fibrodysplasia (myositis) ossificans progressiva. Human Bone Morphogenetic Protein-2 (BMP- 2) is a disulfide-bonded homo-dimeric protein with an apparent molecular weight of 26 kDa. BMP-2 regulates similarly to its nearest homologue BMP-4, diverse fundamental processes during embryonic development. BMP-2 and other BMP proteins have great potential for medical therapeutic applications, in particular because they allow or at least accelerate the ossification of extensive bone lesions. Human recombinant BMP-2 produced in HEK cells is a glycosylated disulfide-linked homodimer, having a molecular weight range of 30-38 kDa due to glycosylation. It has been purified by proprietary chromatographic techniques.

Activity
rHu BMP-2 is fully biologically active when compared to a standard. The specific activity was determined by the dose-dependent induction of alkaline phosphatase production in the ATDC-5 cell line (Mouse chondrogenic cell line) and is typically 20-100 ng/ml.

Formulation
Sterile-filtered, white, lyophilized (freeze-dried) powder. Lyophilized from a concentrated protein solution (1 mg/ml) in 1xPBS.
Reconstitution
Please centrifuge product before opening the vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into water or other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu BMP-2 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
