Description

The ABL1 protein plays a critical role in regulating various cellular processes, including actin binding and remodeling, cell motility, adhesion, receptor endocytosis, and autophagy. It is also involved in DNA damage response, aiding in checkpoint repair after moderate damage and triggering apoptosis following severe damage.

ABL1 is most renowned for its fusion with the BCR gene, resulting in the BCR-ABL1 fusion gene, commonly referred to as the Philadelphia Chromosome, which is a hallmark of chronic myeloid leukemia (CML). However, ABL1 can also form fusions with several other partner genes, expanding its relevance in various leukemias. Notable fusions include:
ETV6-ABL1 in acute lymphoblastic leukemia (ALL), myeloproliferative neoplasms (MPN), and Ph-negative CML
RCSD1-ABL1, SEPQ-ABL1, and ZMIZ1-ABL1 in B-cell ALL
NUP214-ABL1 and EML1-ABL1 in T-cell ALL

These gene fusions typically result from the joining of the 5’ sequences of the partner gene with the 3’ sequences of ABL1. While many ABL1 fusion genes are associated with specific leukemia subtypes, both BCR-ABL1 and ETV6-ABL1 have been identified across multiple disease phenotypes. Research suggests that such non-specific fusions may arise in hematopoietic pluripotent or committed stem cells, leading to diverse leukemia presentations.
