Drosophila Models
Drosophila (D. melanogaster) has gradually become an important tool for the study of Alzheimer's disease due to their short life span, along with the increased availability of techniques and tools which enabled the manipulation of certain gene expression. In addition, well characterized neuroanatomy and phenotypes related to Alzheimer's disease have better explained in Drosophila. Orthologues of the genes related to AD, such as APPL, have been identified in Drosophila and more importantly, the proteolytic activity of γ-secretase was found in both Drosophila and humans. However, there is no formation of Aβ peptides due to the lack of β-secretase activity and sequence differences between APPL and APP. Research has shown that expressing human APP and human β-secretase in fly nervous system produced Aβ deposition and substantially shortened the life span of them. Moreover, the neural toxicity could be revealed by the level of Aβ secreted in Drosophila. In addition, a tauopathy model in Drosophila has been established which can be used in genetic screens for modifiers of tau pathology with obvious pathological phenotypes.
