Abstract

This thesis deals with the development of immunochemical multianalyte microarrays, miniaturized bioanalytical platforms carrying the potential for an area the size of a microscope slide sensitively and specifically detected in parallel by hundreds or thousands of analytes. The theoretical part was compiled research on the current situation in the field of protein microarrays. In the practical part, numerous experiments were carried out trying to find an optimal method of preparing such miniaturized analytical array. Variously chemically activated slides, of our own production and commercial, were used for immobilization of biomolecules. Reagents were dispensed using ink-jet microdeposition. The aim was to get a model system of human serum albumin and a monoclonal antibody to work on this format, and I was partially successful in it. Analysis of produced microarrays was performed by the means of fluorescent detection, using a fluorescence microscope and a fluorescence scanner. Suitability of employed techniques for further work in this field was evaluated.
