Description
Placenta growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family of growth factors. PlGF and VEGF share primary structural as well as limited amino acid sequence homology with the A and B chains of PDGF. All eight cysteine residues involved in intra and interchain disulfides are conserved among these growth factors. As a result of alternative splicing, three PlGF RNAs encoding monomeric human PlGF-1, PlGF-2 and PlGF-3 isoform precursors containing 149, 179 and 219 amino acid residues, respectively, have been described. In normal mouse and rat tissues, only one mouse PlGF mRNA encoding the equivalent of human PlGF-2 has been identified. Mouse PlGF shares 65% amino acid identity with human PlGF2. The gene for PlGF has been mapped to mouse chromosome 12 and human chromosome 14. PlGF binds with highaffinity to Flt1, but not to Flk1/KDR. The recombinant mouse PLGF-2 is a ~40 kDa dimer containing two peptide chains of 132 and 135 amino acid residues.

Activity
Measured by its ability to bind to immobilized rh-sFlt-1 in a functional ELISA. Recombinant mouse PlGF can bind to immobilized rh-sFlt-1 (100 ng/well) with a linear range at 0.5 - 10 ng/ml.

Formulation
Lyophilized from a steril-filtered protein solution in 25 mM Tris, 75 mM NaCl (pH 8.5) with BSA as stabilizer.
Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water, PBS or 50 mM acetic acid to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at - 20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted protein should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
